Agilent 6460 Triple Quadrupole mass spectrometer with Agilent 1290 UHPLC equipped with additional fluorescence and diode array detectors.

This instrument has both ESI and APCI sources and can perform MS/MS product ion scan; MRM; MS/MS neutral loss and precursor ion scan; SIM. (Funded by CFI)

Recent Applications: Targeted screening and quantification of chemical compounds simultaneously for metabolomics and clinical analysis. Recent applications include measurement of deuterated phenylalanine in plasma and tissue for protein synthesis; measurement of glutathione and related sulphydryl metabolite concentrations in plasma and tissues; measurement of choline, betaine, methionine and dimethyl glycine for analysis of 1-C metabolism.

Agilent 7890GC /5975C Gas chromatograph single quadrupole mass spectrometer.

This instrument is equipped with electron impact and positive and negative chemical ionization CI sources, capable of full scan and SIM operation, and can reference the NIST EI library. (Funded by NSERC)

Recent Applications: Quantitative analysis of volatile derivatized compounds. Recent applications includes plasma measurement of 13C-KIC, leucine metabolite for determination of whole body protein turnover, and deuterated glucose for glucose production.

Luminex 100/200 System uses flow cytometry

Measures biomarkers in complex biological samples: protein analysis (cytokines, chemokines, growth factors, hormones, cell signalling proteins, transcription factors and nuclear receptors), gene expression profiling and genotype analysis. Up to 100 selected assays can be conducted simultaneously on 50 uL samples, allowing cost-effective, high throughput in a minimal sample volume. (Funded by NSERC)

Recent Applications: Panels of up to 23 cytokines measured in a single plasma sample in patients undergoing cardiac surgery or liver resection.

Agilent Seahorse Extracellular Flux-24 (XFe24) Analyzer

Agilent Seahorse Extracellular Flux Analyzer (XFe24): The XFe24 is a 24-well metabolic flux analysis instrument that provides quantitative and real time measures of mitochondrial respiration and glycolysis in cultured cells. The system can be utilized to measure changes in mitochondrial bioenergetics and glycolytic flux in almost any adherent cultured cell including cell lines, primary cells, pancreatic islets, and can be adapted for measures using suspension cell lines. The system allows for the measure of all states of mitochondrial respiration and uses fluorometric probes to measure both O2 consumption and glycolysis simultaneously in 24 cell cultures. Oroboros

The Oroboros O2K Fluorespirometer: The O2K system utilizes an electrode-based system for the high-resolution measure of mitochondrial respiration using either suspension cells, muscle fibers, tissue homogenates, or isolated mitochondria. The system is also equipped with a fluorometer that allows for the simultaneous measure of other mitochondrial parameters, such as ROS production, calcium levels, or membrane potential, using commercially available reporters. BioTek Cytation 5 Cell Imaging Multi-Mode Plate Reader

Cytation 5 Cell Imaging Multi-Mode Reader: The CYT5 system is equipped with fluorometric, absorbance, and luminescent detectors to conduct a wide array of plate-based assays. The instrument is equipped with microscopy capabilities (up to 60x) that can allow for imaging of up to 96 wells and for the visualization of real time changes in fluorescence, absorbance, or luminescence in cultured cells or isolated organelles. The system is also plugged into the XFe24 to allow for the correction of respiration and glycolytic flux in the Seahorse with the number of cells per well in the XFe24 plate. (Funded by NSERC)

Cell Culture Facility

Cell culture facility: The new modernized cell culture facility is equipped with a biological safety cabinet, incubators, microscopes, a table-top swing bucket centrifuge, refrigerated microcentrifuge, and automated cell counter for the cultivation of cell lines and primary cells.
